
Our next-generation lentiviral vector production system incorporates advances from our Expression Cassette Optimization (ECO) platform. We have made advances to 3rd generation lentiviral vector systems to increase titer and safety. The region where we insert custom transgenes is flanked by motifs that limit translational readthrough from internal promoter activity and ultra-low homology backbones provide an additional layer of safety. Our team has deep experience in the manufacture of LentET vectors and an efficient, optimized method well established in house. We are able to make the process of ECO-based optimization and inserting into LentET virtually seamless, allowing our clients to rapidly enter our production process with the experience and process already in place.
The foundation of our differentiation is based on internally developed scientific breakthroughs in transgene bioengineering, what we call Expression Cassette Optimization (ECO). ECO uses artificial intelligence (AI), machine learning, and neural networks to optimize nucleic acid sequence of a gene therapy product for efficient expression in the intended target cell type. ECO is a different approach to transgene optimization. We work with the client to discuss their target product profile and can custom tailor solutions to minimize the iterative design process and bring solutions not typically offered by user-facing/self-serve codon optimization services. Our ECO-based tissue-directed optimization not only increases expression in target tissues, it also offers a non-canonical method of reducing expression in off target issues. ECO is currently tailored for mRNA, AAV, and lentiviral therapies.
ECO generates gene expression sequences with enhanced protein expression, increased stability, and improved safety profiles while considering each client’s unique product profile. Additional features incorporated into our ECO platform include vector selection, which tailors the rules of optimization to the delivery system, Open Reading Frame reduction with neural network driven HLA binding potential analysis, which could lower potential for immunogenicity, optional CpG ablation to improve expression, AlphaFold 2.0 enabled protein structure prediction, with the power to use codon choice to reduce protein misfolding, and RNA sculpting to increase RNA stability, thereby prolonging gene expression. We have shown ECO to improve performance over previously codon optimized transgenes from 2 to 8-fold in both in vitro and, more critically, in vivo settings. Combined with our deep expertise in manufacturing lentiviral vectors, Expression provides clearly superior vector designs to the client and can manufacture best-in-class vector transgenes.
